Since the early to mid-20th century, the pharmaceutical industry has continuously relied on the traditional method of bacterial enumeration to support critical decisions related to the bioburden analysis of utility and purified water (PW). This culture-based technique, grounded in decades of scientific validation, has served as the foundation for assessing microbial contamination and ensuring compliance with regulatory standards for water quality in pharmaceutical manufacturing environments.

Initially, the bacterial counting practice was the most accurate and reliable tool for measuring viable microorganisms and met quality needs for years.
